"What are the main points?" would be just one question though but yeah, it's the best one to start with. Another could be "What do I need to know to understand ____________?"
You have to be really curious about the answer or have something else driving you for the answer. If you have a test on Monday, that's good enough to drive you. If you're just bored and are killing time reading a book, it probably won't do anything.
